Troubleshooting getting started — where to start?

Something is not behaving and I am not sure where to begin. What should I check first, and what information would help others help me?

Run wp jetonomy flush-rules (or visit Settings > Permalinks and save) after changing the base slug. The 404s are stale rewrite rules — regenerating them fixes it immediately.
